Man jailed for naked stunt on London statue

(AP) ? A man who climbed naked onto an equestrian statue in London's government district, ripped off its sword and bit it has been sentenced to 12 weeks in jail.

Dan Motrescu brought central London traffic to a standstill when he mounted the bronze statue of the 19th-century Duke of Cambridge on Nov. 23.

Police cordoned off Whitehall, a street that is home to several government departments, as Motrescu climbed up and down the statue, at one point balancing himself on the duke's head.

It took several hours for officers to talk him down.

Motrescu, a Ukrainian citizen of no fixed address, was convicted Tuesday of possession of an offensive weapon, criminal damage and a public order offense. A magistrate at Westminster Magistrates' Court sentenced him to 12 weeks in jail.

"The Honeymooners" musical - because TV land junkies demanded it?

LOS ANGELES (TheWrap.com) - Ralph Kramden's signature threat, "to the moon, Alice," will soon be delivered with musical accompaniment. Satisfying the demands of millions of TV Land junkies, the Old Globe said Thursday that it will kick off its 2013-14 Season with the world premiere of "The Honeymooners," a new musical based on the "Golden Age" television series.

Tony Award winner Michael McGrath ("Nice Work If You Can Get It") has the unenviable task of trying to make audiences forget the (forgive the fat joke!) larger-than-life presence of Jackie Gleason as the much put-upon bus driver Ralph Kramden. Jerry Mitchell, who the Tony-winning choreographer of "La Cage Aux Folles," will direct and choreograph the musical comedy at the San Diego theater.

"The Honeymooners" boasts a book by Dusty Kay and Bill Nuss, music by Stephen Weiner and lyrics by Peter Mills.

The Old Globe said the rest of the creative team and casting will be announced at a later date. The show will run on the Donald and Darlene Shiley Stage in the Old Globe Theatre from September 8 - October 27, 2013. Opening night is slated for Sunday, September 22.

If it connects with critics and audiences, the show could find itself moving cross-country to Broadway. The Old Globe has been the launching pad for a number of musicals, such as "The Full Monty," "Dirty Rotten Scoundrels" and "A Catered Affair" that ultimately migrated to the Great White Way.

"As a kid I was a full-blown ?Honeymooners' addict, and I ?bang-zoomed' away many more hours with the Kramdens and Nortons than I should probably admit," said Old Globe Artistic Director Barry Edelstein. "That's why it's a special joy to see Ralph, Ed, Alice and Trixie come to vivid life on our stage and to introduce these iconic American characters to a new generation."

The show will follow Kramden and his buddy, Ed Norton, after they win a jingle contest and struggle to adjust to life as players on Madison Avenue. If it works, maybe the theatrical community can look forward to musical versions of other classic shows like "F Troop."

Combating MRSA: Researchers study toxin

ScienceDaily (Dec. 4, 2012) ? The Caenorhabditis elegans, a small worm called a nematode, scurrying across a Petri dish has helped lead to discoveries about community-associated MRSA (methicillin-resistant Staphylococcus aureus).

"In the last year this organism has killed more people in the United States than AIDS," said Kathleen Dannelly, an Indiana State University associate professor of biology leading the community-associated MRSA study. "It's going to get worse."

MRSA identifies a staph infection that is unable to be defeated by most penicillin-based drugs. Community-associated MRSA varies from the hospital-related one, which tends to be a more virulent strain picked up by people admitted to hospitals. Community-associated MRSA can cause toxic shock syndrome or blood and bone infections.

"They've picked up this incredible invasiveness mechanism that we don't understand at all," Dannelly said. "It's frightening what it can do."

A normal staph infection looks like a boil and remains in the skin. With MRSA, in the worst strains, the bacteria can eat down through the flesh to the bone in 36 hours.

Dannelly became interested in studying the community-associated MRSA in 2004 when an outbreak infected members of Indiana State University's football team. Players who received cuts and abrasions during games and practice found themselves diagnosed with MRSA and, in some cases, undergoing surgery for it.

Many people unknowingly carry MRSA on their skin as part of their bacteria and remain uninfected as long as they receive no cuts or scrapes to allow the bacteria to enter the body, Dannelly said. Six years ago, Dannelly and her students conducted a blind sidewalk study, swabbing the noses (MRSA thrives in warm, moist places) of 500 students, faculty and staff. They found that 1 ? percent carried MRSA bacteria. Five years later, Dannelly and her researchers again conducted the blind study and they found the number of MRSA carriers had grown to 2 percent of the study group.

"The people had no idea they were carrying it," she said.

In the meantime, the scurrying nematode, which is only a few millimeters long, helped lead researchers to an interesting discovery. By comparing the genomes of community-associated MRSA and the staph infection, they found significant differences to investigate.

"This organism is so invasive we thought there have to be toxins that we don't know about," Dannelly said.

After cloning 15 genes, researchers have, thus far, purified six to determine what the proteins do. Researchers have found that MRSA's different proteins have unique functions such as sticking to human cells or interacting with white blood cells.

One project led researchers to a novel toxin that affects the nematode, which eats bacteria and has an immune system and nervous system in its body making it ideal for tests. They put the worm in the Petri dish with the toxin and watched.

"When you feed it the toxin it runs to the edge of the Petri dish to try to escape," Dannelly said. "This toxin kills the worm in eight minutes. It's really toxic."

Now researchers are testing the toxin on tissue cultures to see how it affects human cells.

As doctoral student Anupama Ramalinga worked in the lab among the tissue cultures, she said the toxin only seems to work on human cells, not on other mammals on which she tested it. She studied tissue cultures under the microscope and counted the number of cells so "in the test I know how many cells my protein killed," she said.

Research into MRSA's affect on humans and its proteins' roles remain important to figuring out the bacteria and how to stop its advance.

"If we don't know our enemy we can't defeat them. If we know our enemy, we can come up with new ways to defeat them," said Ramalinga, from Bangalore, India, who is researching the toxin's effect on tissue cultures. "This type of research will help us understand the bacteria, why it's becoming more robust, stronger."

While Ramalinga finishes her research and works with Dannelly to author a paper on the findings, some common practices can help people fight the bacteria. People can take steps against community-associated MRSA and good personal hygiene is the first step. Also, Dannelly said washing with antimicrobial soap and using a disinfectant in the nose can eliminate the microbes that cause the disease.

"You can disinfect a person and change their flora," she said.

Keep your holiday pay & bonuses legal - Business Management Daily

gift wrapped with moneyDon?t confuse Uncle Sam with Santa Claus. When it comes to violating pay laws around the holidays, the feds won?t be generous if your organization is on the naughty list.

Here are four rules to make sure holiday pay complies with the Fair Labor Standards Act (FLSA) and IRS rules.

Pay for nonexempt employees

Rule #1: Eight hours is the basis. Just as the FLSA doesn?t mandate that you offer paid holidays, it also doesn?t specify how you should calculate pay if you decide to offer the benefit. That means you have lots of latitude.

For example, even if your nonexempt employees usually work 10-hour days, you can pay them for eight holiday hours.

Rule #2: Holiday pay doesn?t count toward overtime. If you do pay for un??worked holidays, and employees work additional hours that week, don?t factor the holiday pay into their regular rates as you calculate overtime pay.

For example, if you offer eight hours of holiday pay and employees also work four 10-hour days, you don?t have to pay OT.

Pay for exempt employees

Holiday pay for exempt employees is trickier, since exempts generally must be paid their full salaries for any week in which they do any work.

Rule #3: You can mandate time off. If you close for a holiday, and you have a bona fide benefits plan, exempts can be required to take those days off, provided they continue to receive their guaranteed salary.

Note: You must pay full salaries to every exempt who performs any work during a workweek, even those who may have run out of accrued leave or who have negative balances in their leave banks.

Rule #4: You don?t need to pay exempts who don?t do any work during a workweek. Thus, if you shut down for the entire week between Christ??mas and New Year?s, you needn?t pay exempts anything. They may use vacation or other accrued time, if they have it.

How to handle holiday bonuses

If you pay holiday bonuses, keep in mind that the IRS always considers them to be taxable income. That means you must withhold both income taxes and FICA taxes. But how bonuses are paid and taxed?in separate checks or combined with regular pay in one check?is up to you.

Rule: Provided you withhold income taxes from employees? regular pay, you may pay bonuses in separate checks and withhold income taxes at the flat 25% supplemental withholding rate.

Naturally, that?s going to reduce the amount of the bonus employees can actually spend. What can you do to ensure your employees receive the full bonus amount? Gross-up the bonus to cover the income and FICA taxes due.

Let?s say you want your employees to pocket a full $500 bonus. To gross-up, subtract the flat 25% income tax withholding rate and the employee FICA tax rate from 1:

$500 ? (1 ? 0.25 ? 0.0565) = $720.98

The bonus amount would be $720.98. You would withhold $220.98 in income and FICA taxes, leaving the employee a take-home bonus amount of $500.

Trouble. Big trouble. Big luscious lips and deep sensual eyes, staring at me. Big, deep and up-to-my-neck-in-it trouble.

Cath is due back any minute. Enough time to contrive an apology, but not enough to undo this mess.

"Don't spend all morning surfing dodgy sites." She winked as she headed for the door. "You need to buy milk and something for dinner. Speaking of surfing, don't forget to renew the firewall and anti-virus subscription; it expired yesterday. I'll be back at two to print out my portfolio."

I should have got my act together and headed out to the shops immediately, renewing the subscription on my return, rewarding myself with a coffee. The rest of the day would have been mine to squander. Should have... but as the door closed, my subconscious had already prioritised surfing with coffee over shopping and subscription renewal.

OK, I'll admit I may have looked at some sites that had nothing to do with my thesis write-up... including a couple that didn't involve pictures of cats. I was tempted to renew the subscription as further procrastination, but it was midday and the high street would be busy, getting busier.

Well, I've just renewed the sub and scanned and fixed the PC, but that's locking the barn door after the horse has bolted and the printer cartridges have emptied. How was I to know one of those sites had the S3xD0ll virus?

When I got back from the shops I thought Cath had returned early because the printer was chuntering away in the background. Cheap 3D printers have knocked the low end out of the consumer products market, with open-sourced and pirated designs online further squeezing the product designer jobs market. Cath, however, has secured an interview and she was going to print out some of her work to take along. In preparation she'd bought litres of plastic and metal powders... now used up. In their place I have a life-sized animatronic sex doll to explain away. Big luscious lips and deep sensual eyes, staring at me with preprogrammed expectation.

And that's the front door.

Curiosity finds carbon ? but is it from Mars?

A tantalising whiff of carbon-based compounds has been picked up by NASA's Curiosity Mars rover. But it's still not clear whether the eagerly awaited results show the true chemical nature of Mars or are the products of Earthly contaminants.

One of the main goals of the NASA rover is to search for signs of Martian habitability, including organics ? carbon-containing compounds that are the building blocks of life. The rover's first meals of baked Martian soil showed carbon and hydrogen reacting with chlorine inside the robot's ovens, creating organic molecules.

This taste of carbon is intriguing but it's a far cry from recent feverish speculation that the rover found definitive evidence for organics on the Red Planet. While the chlorine is almost certainly from Mars, it is still unclear whether the carbon is native Martian material or something from Earth that was trapped in the rover. Even if the carbon is from Mars, it may come from inorganic sources, like carbonate rocks.

"The rover has made this detection of simple organic compounds," said project scientist John Grotzinger of NASA's Jet Propulsion Laboratory in Pasadena, California. "We just don't know if they're indigenous to Mars or not."

Chemical diversity

The result is only one of the intriguing chemical discoveries the NASA rover has pulled from a wind-blown patch of sand at a site called Rocknest, unveiled today at the American Geophysical Union (AGU) 2012 fall meeting in San Francisco.

Since it landed in Gale Crater in August, Curiosity has been firing up its science tools one by one. Star of the show at the AGU meeting was the Sample Analysis at Mars (SAM) experiment, which can sniff the Martian atmosphere, and can analyse gases given off when soil samples are heated to as much as 1100 ?C.

SAM was the last major instrument to come online. Excitement surrounding the consistency of results emerging from multiple samples was what prompted Grotzinger to talk about data that's "for the history books" in a US radio interview almost two weeks ago, sparking a media frenzy.

"These results are an unprecedented look at the chemical diversity in the area," Michael Meyer, NASA's lead scientist for Mars exploration based in Washington, DC, said today at an AGU press briefing.

Earthly trace

SAM has been digesting scoops of fine sand, with grains less than 150 micrometres across. "It's finer than sugar, but coarser than something like flour," explains Ken Edgett, principal investigator for the rover's Mars Hand Lens Imager, which has taken detailed pictures of the material. It is this sand that revealed the chlorinated compounds.

SAM took several scoops from the Rocknest site in early October, shaking its tray and dumping out the first few loads to clean the instrument and make sure any Earthly molecules were flushed out of its system. The fifth scoop was delivered to SAM's oven, which heated it until the compounds inside released their characteristic gases.

The results revealed chlorinated carbon compounds, which probably formed inside SAM's oven rather than being picked up directly from Mars. The question is where the carbon comes from. Even after a thorough rinse, it's possible the SAM oven still contains traces of Earthly carbon.

If it's Martian in origin, the carbon may be from organic hydrocarbons or inorganic carbonates. The chlorine, meanwhile, may be from chemicals called perchlorates, which were previously spotted by NASA's Phoenix lander near the Martian north pole. These compounds are found in rocket fuel and other explosives and are typically considered toxic, but some extreme microbes on Earth may use them as an energy source.

?Hold the show'

The team will compare the result to tests of a sample of carbon-based material brought from Earth, Paul Mahaffy, SAM's principal investigator, said at the AGU meeting. "If we see the same stuff that we thought might be from Mars, we've got to say, ?Hold the show, this might be terrestrial stuff'."

Because the sand at Rocknest is representative of material blown from around the Martian surface, the new results provide a solid baseline describing the planet's global conditions. The data will later be compared to layers of rock at the rover's ultimate destination ? a 5-kilometre tall mountain known as Aeolis Mons, sometimes called Mount Sharp. Some of these layers formed more than 3 billion years ago, when it's thought that Mars was warm, wet and may have supported life.

Curiosity's sampling of the Martian soil has also revealed that the ratio of the heavy isotope deuterium to ordinary hydrogen is five times higher on Mars than on Earth ? a discovery that may help pinpoint when and how the Red Planet lost most of its formerly thick atmosphere.

Curiosity is now getting ready for its long trek to Aeolis Mons, taking in other points of interest on the way. Grotzinger likens Curiosity to a car with a 10,000-page user manual, which was still being written as the science team tested its instruments. With all the gear now up and running, it's time to drive, he says. "Our car is ready to go."

The Chinese stock market has been the worst major market in the world this year (see right).

Down again today, the Shanghai Composite index is trading near a four-year low.

Even though the Chinese economy has had its bumps, the growth rate continues to be staggering, and most recent economic data show growth is accelerating again.

So why have the stocks done so poorly relative to the economy?

The most asked question from global investors is why Chinese onshore A-shares did not respond to the improving fundamentals. Is this because these macro indicators have low quality and give us a wrong picture? We don?t think so. Very often the sentiment-driven A-share market is a barometer for Chinese investors? mass psychology instead of the real economy. More specifically, domestic investors might get increasingly disappointed about prospects of future financial reforms, especially reforms on stock markets. Availability of other investment channels such as wealth management products and housing also divert demand. Heavy-handed interventions in the past year sows the seed of bear market now as about 100 IPOs have been approved but yet to be listed and 700 more are in the pipeline.

In other words, stocks aren't the economy.? It's a reflection of psychology, which is arguably the case for stocks everywhere.

Go with the flow in flood prediction

ScienceDaily (Dec. 3, 2012) ? Floods have once again wreaked havoc across the country and climate scientists and meteorologists suggest that the problem is only going to get worse with wetter winters and rivers bursting their banks becoming the norm. A team based at Newcastle University and their colleagues in China have developed a computer model that can work out how the flood flow will develop and where flooding will be worst based on an understanding of fluid dynamics and the underlying topology of a region.

Writing in the journal Progress in Computational Fluid Dynamics, Newcastle civil engineer Qiuhua Liang and colleagues and Chi Zhang of Dalian University of Technology and Junxian Yin, China Institute of Water Resources and Hydropower Research in Beijing, explain how they have developed an adaptive computer model that could provide accurate and efficient predictions about the flow of water as a flood occurs. Such a model might provide environmental agencies and authorities with a more precise early-warning system for residents and businesses in a region at risk of flood. It could also be used by insurance companies to determine the relative risk of different areas within a given region and so make their underwriting of the risk economically viable.

The model is based on a numerical solution to the hydrodynamic equations of fluid flow . This allows the researchers to plot the likely movement of water during a dam break or flash flood over different kinds of terrain and around obstacles even when flood waves are spreading rapidly. The researchers have successfully tested their model on real-world flood data.

The team points out that flood disasters have become a major threat to human lives and assets. "Flood management is therefore an important task for different levels of governments and authorities in many countries?, the researchers explain. "The availability of accurate and efficient flood modelling tools is vital to assist engineers and managers charged with flood risk assessment, prevention and alleviation."

Germany's Merck Serono to produce medicines in UAE

ABU DHABI (Reuters) - Germany's Merck Serono said it would team up with an Abu Dhabi firm to produce medicines for the domestic and regional markets, the first multinational of its kind to make branded products in the United Arab Emirates.

Merck Serono, the pharmaceutical arm of Merck KGaA, will initially produce two of its products at Neopharma's facilities in Abu Dhabi, the German firm's first such partnership in the Middle East, the company's CEO said.

"The Middle East is very important and the fastest growing region for our products," Stefan Oschmann told reporters. "We plan to meet 100 percent of the regional demand for the products," he added.

Oschmann declined to give specific figures but said revenues from emerging markets accounted for a third of Merck's total.

The two products to be produced at Neopharma's facilities are Euthyrox, a synthetic thyroid hormone and Glucophage, for people suffering from type 2 diabetes.

Production of both branded products will start in 2013.

The UAE suffers from a high prevalence of diabetes. Some 827,000 people between the ages of 20 and 79 have diabetes in the UAE, according to the Ministry of Health.

Treatment of diabetes accounts for about 40 percent of the UAE's overall healthcare expenditure, Amin al Amiri, assistant under-secretary for Medical practice & license at the ministry of health said.

"This alliance will provide increased supply of trusted branded medicines needed to help ease the diabetes crisis which is straining the financial resources of the UAE," he said.

The Middle East is the largest market for Merck's diabetes drug, Glucophage, which is currently imported from Europe.

Hi, I am Phil. I was made redundant (retrenched) from my IT job in England in August 2010. We decided to relocate to Singapore where we have more relatives.

My wife started her job in Childcare in March 2011. My role as stay-at-home dad started in Singapore, when Zephany was about 9 months old and Zack was 5 and started his K2. Before I was committed the role as a stay-at-home dad, I admit that like most typical men, leaving the care of children to my wife.

When I was working in England, I worked shift, either from 7am to 4pm or 8am to 5pm or 2pm to 10pm. When I came home from work, if I was early enough for the kids to be awake. I used to ask ?how was your day? and ?how was school?. Then I played with the kids till bedtime, read a book and put them to sleep.

When I started to be a stay-at-home dad, I soon learned new set of skills, became juggler of 2 different age children?s needs, from trips to the park, feeding them, changing nappies, putting them to sleep, taking Zack to school. In the morning, bring Zephany along to pick Zack up from school at lunch time and in the afternoon, spent time putting Zephany to sleep and playing with Zack. Sometimes I get asked on the street, ?where is their mum?, I will reply, ?she?s at work? and I was also asked ?who is looking after the kids?. I think people expect me to have domestic helper or mother-in-law to look after my children. I told them that I look after them and they are mine to look after. People also seem surprise that a man of 50s can do this or maybe want to do it. Sometimes people think that I am the grandfather, people are not used of seeing an older dad, I am very proud of my kids, especially when Zack said to them that ?he?s my dad?.

As stay-at-home dad, I sometimes feel undervalued and lonely during the day where people rushing off to work, I sometimes get jealous of them. I feel left out of life sometimes. Miss the journey to work, chat with colleagues about football, about last night?s TV programs, we moan about going to work, coffee break, miss the ?wall to kick at? attitude, missing the day to day life in the office. I miss working life. Some people said to me ?you are lucky, not going to work?. Yes I am! But people fail to understand that I miss out on things they take for granted. My role in life has changed now that I appreciate how hardworking looking after children can be.

Now that my daughter goes to childcare and my son in primary school, I feel more undervalued and lonelier. In the morning, when my wife start work early, I bring Zephany to childcare then bring Zack for breakfast or to the park before going to school as he is in the afternoon session. Then I will bring Zack to school in the afternoon. My day is full of getting him ready for school each day, taking him and picking him up from school, making sure he done his homework and checking his completed work. I am all by myself in the afternoon, doing some housework, looking for jobs on the website, etc. I feel useless sometimes, when my wife working hard and I am doing nothing at home. I miss working.

During school holidays, I bring Zack for bike rides, trips to Science centre, swimming, etc.

For those who are reading this and wonder why I can?t work, is because I am holding a Long Term Visit Pass (LTVP), I am not allowed to work or companies won?t want to employ me, unless I have a Long Term Visit Pass Plus (LTVP+) or a PR. Some of our friends asked, why can?t I get a PR, my wife and children are Singapore Citizens. We leave that for the government to tell us. We do feel left out and neglected by the government sometimes, where most foreigners who work and live in Singapore get PR so easily and they have no connection with Singapore but my wife and children and also my in-laws are Singapore citizen but the government can?t help us even after few appeals.

Anyway, I enjoy being a stay-at-home dad and am also desperately looking for a job. When I am home alone in the afternoon, I am always looking forward for my wife and daughter to come home. I will meet them at the shops. I look forward to every evening when I sit together with my wife and children by the dining table having dinner, talking about the day at work and in school. That?s the best part of the day.

Well, I can tell you, I have been busy! It?s been frustrating, stressful, enlightening, exciting, fun and fulfilling time. Learned a lot being a father and lots about myself. An adventure that I wouldn?t want to miss out on. I wonder will people understand that it?s not been a holiday, I have been doing an important job where some people might want to but not able to. Being a parent is the most important job. Most fathers don?t spend enough time with their children.

Humans are weird, when we are jobless, we wish to work, when we are working, we wish to be at home. That?s life!

Personally, I do not know of any SAHDs and I'm glad Phil took the time to share his story on the blog today.? We often hear how SAHMs feel undervalued, so it's interesting to hear from the other side and learn that the same frustrations uttered by SAHMs and discussed by guys in the same position too. A recent study found that SAHDs has doubled in the past decade in the U.S.,? and that these daddies are happy in their role. They love what they do and felt pride in being more involved with their kids' parenting. Gender roles are blurring as society evolves, and no matter who brings home the bacon, let's not forget to take the time to thank one another for sacrifices made. Because ultimately, it's all for the kids.

Childhood trauma leaves mark on DNA of some victims: Gene-environment interaction causes lifelong dysregulation of stress hormones

ScienceDaily (Dec. 2, 2012) ? Abused children are at high risk of anxiety and mood disorders, as traumatic experience induces lasting changes to their gene regulation. Scientists from the Max Planck Institute of Psychiatry in Munich have now documented for the first time that genetic variants of the FKBP5 gene can influence epigenetic alterations in this gene induced by early trauma.

In individuals with a genetic predisposition, trauma causes long-term changes in DNA methylation leading to a lasting dysregulation of the stress hormone system. As a result, those affected find themselves less able to cope with stressful situations throughout their lives, frequently leading to depression, post-traumatic stress disorder or anxiety disorders in adulthood. Doctors and scientists hope these discoveries will yield new treatment strategies tailored to individual patients, as well as increased public awareness of the importance of protecting children from trauma and its consequences.

Many human illnesses arise from the interaction of individual genes and environmental influences. Traumatic events, especially in childhood, constitute high risk factors for the emergence of psychiatric illnesses in later life. However, whether early stress actually leads to a psychiatric disorder depends largely on his or her genetic predisposition.

Research Group Leader Elisabeth Binder of the Max Planck Institute of Psychiatry examined the DNA of almost 2000 Afro-Americans who had been repeatedly and severely traumatised as adults or in childhood. One-third of trauma victims had become ill and was now suffering from post-traumatic stress disorder. The risk of developing post-traumatic stress disorder rose with increasing severity of abuse only in the carriers of a specific genetic variant in the FKBP5 gene. FKPB5 determines how effectively the organism can react to stress, and by this regulates the entire stress hormone system. The scientists hoped to cast light on the mechanisms of this gene-environment interaction by comparing modifications of the DNA sequence of victims who had not become ill with that of those who had.

The Munich-based Max Planck scientists were then able to demonstrate that the genetic FKBP5 variant does make a physiological difference to those affected, also in nerve cells. Extreme stress and the associated high concentrations of stress hormones bring about what is called an epigenetic change. A methyl group is broken off the DNA at this point, causing a marked increase in FKBP5 activity. This lasting epigenetic change is generated primarily through childhood traumatisation. Consequently, no disease-related demethylation of the FKBP5 gene was detected in participants who were traumatised in adulthood only.

Torsten Klengel, a scientist at the Max Planck Institute of Psychiatry, explains the findings of the study as follows: "Depending on genetic predisposition, childhood trauma can leave permanent epigenetic marks on the DNA, further de-repressing FKBP5 transcription. The consequence is a permanent dysregulation of the victim's stress hormone system, which can ultimately lead to psychiatric illness. Decisive for victims of childhood abuse, however, is that the stress-induced epigenetic changes can only occur if their DNA has a specific sequence."

This recent study improves our understanding of psychiatric illnesses which arise from the interaction of environmental and genetic factors. The results will help tailor treatment particularly for patients who were exposed to trauma in early childhood, thereby greatly increasing their risk of illness.

Genome-scale study identifies hundreds of potential drug targets for Huntington's disease

Scientists searching for ways to develop treatments for Huntington's disease (HD) just got a roadmap that could dramatically speed their discovery process. Researchers at the Buck Institute have used RNA interference (RNAi) technology to identify hundreds of "druggable" molecular targets linked to the toxicity associated with the devastating, ultimately fatal disease. The results from this unprecedented genome-scale screen in a human cell model of HD are published in the November 29, 2012 edition of PLOS Genetics. The work was is a collaboration between Buck Institute faculty members Robert E. Hughes, Ph.D., Sean Mooney, Ph.D., Lisa Ellerby, Ph.D. and Juan Botas, Ph.D. at the Baylor College of Medicine.

HD is a devastating and incurable progressive neurodegenerative genetic disorder that affects motor coordination and leads to severe physical and cognitive decline. Currently, there are about 30,000 people in North America diagnosed with HD and another 150,000 people at risk for developing the disease. The disease pathology stems from a mutation in the huntingtin gene (HTT), resulting in the accumulation of a toxic protein leading to neuronal cell death and systemic dysfunction. Buck Scientists screened more than 7,800 genes pre-selected as potential drug targets to identify modifiers of HD toxicity in human cells, using technology that silences specific genes prior to analysis.

Lead author Robert Hughes said that among the diverse range of modifiers identified, this study showed that RRAS, a gene involved in cell motility and neuronal development, is a potent modulator of HD toxicity in multiple HD models. "Our data indicates that the pathogenic effects of the HTT mutation on this pathway can be corrected at multiple intervention points and that pharmacological manipulation of RRAS signaling may confer therapeutic benefit in HD," Hughes said. Follow up work on the RRAS pathway is now underway in the Hughes lab and in the lab of Buck faculty member Lisa M. Ellerby, PhD.

Hughes said many molecular hits identified in the screening were validated in human cell, mouse cell and fruit fly models of HD ? and that all the data from the study will be available to the public. "Our hope is that HD researchers will look at these targets and find modifiers relevant to the areas they already work on," said Hughes. "Ideally, pharmaceutical companies already working on some these pathways could build on their current knowledge and expertise by focusing their attention on the challenge to develop therapies for HD."

(AP) ? The world's first nuclear-powered aircraft carrier was retired from active service on Saturday, temporarily reducing the number of carriers in the U.S. fleet to 10 until 2015.

The USS Enterprise ended its notable 51-year career during a ceremony at its home port at Naval Station Norfolk, where thousands of former crew members, ship builders and their families lined a pier to bid farewell to one of the most decorated ships in the Navy.

"It'll be a special memory. The tour yesterday was a highlight of the last 20 years of my life. I've missed the Enterprise since every day I walked off of it," said Kirk McDonnell, a former interior communications electrician aboard the ship from 1983 to 1987 who now lives in Highmore, S.D.

The Enterprise was the largest ship in the world at the time it was built, earning the nickname "Big E." "It's shown that the aircraft carrier can evolve as a platform with many payloads relevant for five decades and will be part of our national security for the foreseeable future as we bring on the Gerald Ford to replace the Enterprise."

The Gerald R. Ford will be the first of a new class of aircraft carriers, but it will be several more years before it joins the fleet. It will eventually be towed to Washington state for scrapping.

The nuclear-powered aircraft carrier was the eighth U.S. ship to bear the name Enterprise, but it won't be the last. Navy Secretary Ray Mabus said in a video message that a future aircraft carrier would be named USS Enterprise, after the delivery of the USS Gerald R. Ford and the USS John F. Kennedy.

Mabus' announcement drew a standing ovation from those on hand at Saturday's ceremony. Current and former crew members have lobbied heavily to preserve Enterprise's name so its legacy will live on.

"It just seems to be a neat name for a ship. It's better than being named for a politician," said Larry Kosnopfal, one of the ship's original crew members, who now lives in Chadfield, Minn.

When the future USS Enterprise joins the fleet, its commanding officer will be handed a 200-pound time capsule filled with Enterprise memorabilia that includes notes from sailors, insignia and small pieces of the ship. The time capsule was delivered to Greenert for safekeeping until that future commanding officer is chosen.

"Of course, employers are looking for people who have the baseline skills to effectively do the job," Rivera said. "But, beyond that, employers really want people who they will bond with, who they will feel good around, who will be their friend and maybe even their romantic partner."

The study is based on 120 interviews with professionals involved in undergraduate and graduate hiring at elite U.S. investment banks, law firms and management consulting firms. Rivera found that evaluators often valued their personal feelings of comfort, validation and excitement over identifying job candidates with superior cognitive or technical skills.

More than half of the evaluators in the study ranked cultural fit ? the perceived similarity to a firm's existing employee base in leisure pursuits, background ?and self-presentation ? as the most important part of the job interview process.

Rivera did caution, however, that the results don't mean employers are hiring unqualified people.

"My findings demonstrate that ? in many respects ? employers hire in a manner more closely resembling the choice of friends or romantic partners than how one might expect employers to select new workers," she said.

The study also found that the cultural similarities valued at elite professional service firms have critical socioeconomic dimensions.

"Evaluators are predominately white, Ivy League-educated, upper-middle or upper-class men and women who tend to have more stereotypically masculine leisure pursuits and favor extracurricular activities associated with people of their background," Rivera said. "The types of cultural similarities valued in elite firms' ?hiring processes have the potential to create inequalities in access to elite jobs based on parental socioeconomic status."

Rivera believes that while the research only examined elite professional service firms, the results do have implications for occupations across all industries.

"I think that the process is generalizable, but I think the degree to which cultural similarity matters in the decision to hire varies across occupations depending on their technical demands, their degree of social demands, and how structured interviews are," Rivera said. "So, for example, if you were hiring a neurosurgeon, I think there would be more of an emphasis on performance than cultural fit."

In addition, the types of cultural similarities employers value may not be the same for all occupations.

"Cultivating leisure time is a hallmark of the upper middle and upper classes, and it's really important as a class marker and as a source of identity," Rivera said. "You may see different types of cultural similarities that matter in occupations that are less elite."

The study, "Hiring as Cultural Matching: The Case of Elite Professional Service Firms," appears in the December issue of the American Sociological Review.
